Oryza RNA Heat Structome

This project is to study the heat effect on RNA structure change in rice. The wild-type rice were grown in greenhouse and shoot tissue of two-week-old plant was used for library preparation. 1 g of shoot tissue were cut off the plant right before each treatment. After following our previous pipeline of Structure-seq for +DMS treatment, the -DMS sample was processed with no DMS applied as the control. For the heat parallel experiment, -DMS and +DMS samples were prepared. Library preparation was following our previous library construction pipeline with some major changes and optimization. All 12 individual biological libraries was done with unique Illumina TruSeq barcode and been sequenced by HiSeq2500.
